Activités cytotoxiques et pro-oxydantes d'acides gras à très longue chaîne sur des oligodendrocytes murins sauvages et déficients en Abcd1 et Acox1 :…

2010

X-ALD and P-NALD are two rare, peroxisomal metabolic and neurodegenerative diseases. ABCD1 and ACOX1 are known to be responsible for X-ALD and P-NALD, respectively. The actively demyelinating lesions in CNS, exhibited signs of oxidative stress and cell death. The accumulation of VLCFA in plasma and tissue is the biochemical common hallmark to both diseases. First, we characterized a murine oligodendrocytes cell line 158N to use it as a model. This 158N cell line which has characteristics of mature oligodendrocytes (expression of myelin proteins MOG, MBP, PLP), has also functional peroxisomes with Abcd1 and Acox1 proteins. Then, we studied the cytotoxic and pro-oxidative effects of VLCFA (C2…

Caractérisation des figures myéliniques associées à l'accumulation de lipides polaires induites par différents oxystérols cytotoxiques identifiés dan…

2006

Atherosclerosis is a complex and chronic arterial process which is characterized by a remodeling of the vascular wall, associated with inflammatory reactions, proliferation and cell death process, and with accumulation of oxidized lipids among which oxysterols (cholesterol oxidation products) which might play key roles in the initiation and development of atheromatous lesions. Our work performed on U937 and THP1 promonocytic cells, rat aorta embryonic A7R5 cells, and breast carcinoma MCF7 cells (caspase-3 deficient). Different oxysterols, present in large quantity in atheromatous lesions, were used: 7-cétocholestérol (7KC), 7Β-hydroxycholestérol, 25-hydroxycholestérol, cholestérol-5Α, 6Α-ep…

Des canaux Ioniques de la membrane plasmique lors de la mort cellulaire programmée induite par l’ozone chez A. thaliana

2011

Tropospheric ozone is a major secondary pollutant. In addition to its role in greenhouse effect gas, ozone is one of the most toxic air pollutants, and its pollution affects both human health and crop productivity. The work presented in this thesis concerns the role of ion channels in the plasma membrane in response to acute exposure to ozone and their interactions with signaling events leading to O3-induced PCD in A. Thaliana cultured cells. We have shown that cell death was genetically controlled and characterized by cell shrinkage similar to the mechanism of "Apoptosis Volume Decrease" (AVD) described in animal. This process is initially promoted by an early activation of a plasma membra…

Type-2 histones deacetylases and cryptogein-induced cell death in tabacco

2011

Cryptogein, which is secreted by the oomycete Phytophthora cryptogea, is a proteinaceous elicitor of plant defense reactions that activates a set of signaling events leading to the hypersensitive response and to systemic acquired resistance. Although the early cytosolic signaling events induced by cryptogein are well described, the only nuclear events characterized to date are the variations in free calcium concentrations and defense-related gene expression. The characterization of the activation of cytosolic protein kinases, including WIPK and SIPK, by phosphorylation in response to cryptogein highlights the key-role played by posttranslational modifications in cryptogein-induced signaling…

Cellular metabolic alteration : the voice of monoinsaturated fatty acid as therapeutic target

2010

Stearoyl Co-A desaturase (SCD) is the key enzyme of the metabolism of mono-unsaturated fatty acids (MUFA). Its activity  9 desaturase introduces a double bond cis in position 9 of saturated fatty acids (SFA), induced formation of MUFA. Impaired biosynthesis of MUFA is involved in many diseases such as cancer and cardiovascular disease. Cancer cells have a de novo synthesis of fatty acids increased with an accumulation MUFA. This change in the metabolism of fatty acids is associated with overexpression SCD1 which catalyzes the conversion of saturated fatty acids, monounsaturated fat. Several reports have shown that inhibition of SCD1 leads to blockage of proliferation and induction of apopt…
